Where to position a safe really depends on the type of safe and the environment it is in.

The majority of safes will need to be bolted to the floor or wall so it is better to position it where the floor is solid concrete rather than timber as it is more secure.

It is possible to secure the safe to a timber joist if it is a timber floor and not concrete but this needs to be confirmed with your insurance provider.  This also determines where the safe is positioned because the fixing hole needs to be above a joist.

Consideration to the weight of a safe will determine if the upper floor can support its weight. If you require a high security safe that weighs over 400kg then you need to make sure the floor can support its weight.

A hidden safe will obviously need to be positioned where it is most unlikely to be found but still enables the owner easy access.
